Formulation ratios of cryopreservation solutions for different cell types (precautions for preparing cryopreservation solutions)
Cell cryopreservation solution is a specialized formulation used to maintain cells under low-temperature conditions, with the primary goal of protecting them from damage during freezing and thawing. Such solutions typically contain both osmotic and non‑osmotic cryoprotectants, such as dimethyl sulfoxide (DMSO) and glycerol. These agents can penetrate the cell membrane, reducing intracellular and extracellular electrolyte concentrations and preventing the efflux of intracellular water, thereby minimizing ice crystal formation.

Overview of Cell Cryopreservation Solution Preparation
The commonly used cryopreservation solution is formulated as culture medium : serum : DMSO = 7:2:1, which is suitable for most cell lines and ensures a high post‑cryopreservation viability. For cell lines that require long‑term storage or are particularly valuable, the serum:DMSO ratio should be adjusted to 9:1, providing enhanced protection and helping to maintain their desirable biological characteristics after thawing.
Different cell cryopreservation solution formulation ratios
In fact, most cell types do not require 20% or 90% serum; a complete culture medium containing 10% serum can be used to prepare a 5% DMSO–containing solution for cryopreservation, and some even do not need serum at all.
Some precautions for preparing cryopreservation solution
✔ Select an appropriate cell state: Cells in the logarithmic growth phase with a confluence of approximately 80%–90% should be chosen for cryopreservation to ensure that the cells are in optimal condition at the time of freezing.
✔ Cell density: The cell density during cryopreservation should be appropriate; a recommended range is 3 × 10^6 to 1 × 10^7 cells/mL. A cell density that is too low may compromise viability.
✔ Avoid prolonged exposure at room temperature: After adding cryopreservation solution to the cell suspension, minimize the time it spends at room temperature, as DMSO can cause significant cellular damage under these conditions.
✔ Use high-quality cryovials: Ensure that you use sterile, cryoprotective‑resistant vials, and clearly label them during aliquoting for easy identification later.
✔ Serum-to-DMSO ratio: A common ratio is 90% serum + 10% DMSO, or 55% basal medium + 40% serum + 5% DMSO; adjust according to the specific cell type and experimental requirements.
