Why is the reproducibility of CCK-8 assay data poor?
Publication date:2026-10-29
Page views:
1. The reagents in the outermost ring of the microplate are prone to volatilization; it is recommended to fill only the outermost wells with culture medium and not use them for assay purposes.
2. CCK‑8 may adhere to the well walls; after adding CCK‑8, gently tap the plate to promote thorough mixing.
3. If the cell density is too high or too low, it is advisable to pre‑optimize the conditions within a range of 1,000–100,000 cells per well.
4. Air bubbles in the wells can interfere with ELISA readings; ensure that each well is free of bubbles before measurement.
