Can primary cells be cryopreserved? What are the reasons for the low viability after thawing and recovery?
This depends on the cell type. Certain cell types, such as neurons, glial cells, and some slowly proliferating epithelial cells, are not recommended for expansion and subsequent cryopreservation. Other cell types, including fibroblasts, astrocytes, renal mesangial cells, and astroglial cells, can be expanded and cryopreserved. However, it should be noted that the cryopreservation process may alter cellular growth characteristics. Low post‑thaw viability warrants consideration of pre‑cryopreservation cell viability, cell density, and the cryopreservation medium—whether a serum‑containing cryoprotective solution or a one‑step cryopreservation formulation—as different methods require distinct handling protocols.
