IF=15 | A team from Zhejiang University has developed M1 macrophage–derived fusion nanovesicles, enabling synergistic tumor‑killing through a triad of sonodynamic therapy, ferroptosis, and immunotherapy.
I. Literature Review
Recently, Hu Yihé and Xie Jie, chief physicians, along with Researcher Ma Tianliang from the Department of Orthopedics at the First Affiliated Hospital of Zhejiang University School of Medicine, jointly published an academic paper titled “M1 macrophage-derived fusogenic nanovesicles co-loaded with Fe3O4/IR780 suppress osteosarcoma via a sonodynamic-ferroptosis-immune triad” in the Journal of Nanobiotechnology (latest impact factor: 15).
This study introduces a multifunctional biomimetic nanoplatform that exerts its therapeutic effects through the synergistic interplay of ultrasound‑mediated photothermal therapy, ferroptosis, and immune activation, offering a highly promising and effective combinatorial strategy for the treatment of osteosarcoma.

II. Literature Summary
Osteosarcoma is a highly aggressive bone malignancy with a strong propensity for metastasis. Current treatment modalities are insufficient to effectively control its progression, resulting in suboptimal clinical outcomes. Recent research suggests that inducing ferroptosis and activating antitumor immune responses may represent promising therapeutic strategies. To this end, we have engineered a biomimetic nanoplatform—IR780@NVEV-Fe3O4—by integrating extracellular vesicles derived from M1‑type macrophages with nano‑vesicles preloaded with Fe3O4 nanoparticles and the ultrasound‑sensitizing agent IR780. This platform aims to synergistically combine sonodynamic therapy, ferroptosis induction, and immunotherapy into a unified approach. The IR780@NVEV-Fe3O4 nanoplatform exhibits exceptional tumor‑targeting capabilities and responds robustly to ultrasonic stimulation.
In vitro, the ultrasound‑activated nanoplatform effectively induces ferroptosis in osteosarcoma cells, characterized by exacerbated lipid peroxidation, impaired mitochondrial function, and suppression of GPX4/xCT axis activity. Notably, this process also triggers immunogenic cell death, thereby promoting dendritic cell maturation and the polarization of macrophages toward an M1 phenotype. In murine osteosarcoma models, the combined use of this nanoplatform and ultrasound markedly inhibits subcutaneous tumor growth and pulmonary metastasis. More importantly, when this sonodynamic therapy is paired with an anti‑PD‑L1 antibody, it elicits a robust systemic antitumor immune response, as evidenced by increased CD8+ T‑cell infiltration within the tumor microenvironment. ⁺ T Increased cellular infiltration, the establishment of long-term immunological memory, and a significant prolongation of host survival.
Overall, this study presents a multifunctional biomimetic nanoplatform that exerts its therapeutic effects through the synergistic interplay of “ultrasound‑induced therapy, ferroptosis, and immune activation,” offering a highly promising and effective combinatorial strategy for the treatment of osteosarcoma.

III. Quote product
In this study, ten products from Li Ji Bio® fulfilled four core functions—basic buffering (PBS/TBS), cell culture (DMEM/antibiotics/EZ antibacterial agent), functional assays (CCK‑8/BSA/4% PFA), and protein analysis (rapid blocking solution/chemiluminescent substrate)—covering the entire experimental workflow from cell culture to nanoparticle formulation, in vitro and in vivo functional validation, and molecular mechanism elucidation. These reagents served as critical support for generating the data presented in the manuscript.
