PBS can provide a relatively stable ionic environment and pH‑buffering capacity, typically serving as a solvent to dissolve and protect reagents. It is a commonly used buffer salt solution in biology, employed in applications such as molecular cloning. DPBS is a balanced salt solution designed for short-term maintenance of cell viability, preserving the structural and physiological integrity of isolated cells within a limited time frame. With a pH of 7.2–7.4—close to that of human body fluids—and an osmotic pressure equivalent to blood, it contains no Ca²⁺ or Mg²⁺ ions. DPBS is suitable for various cell culture applications, including pre‑dissociation cell washing, transport of cells or tissues, and dilution of cells for counting and reagent preparation. In fact, PBS is a general term encompassing a family of phosphate‑buffered solutions; while there are standard formulations, the term is not restricted to any single one. DPBS, on the other hand, was originally developed by Renato Dulbecco as a customized PBS formulation tailored to his experimental needs. Due to its widespread use, the name has become relatively standardized over time. Essentially, both DPBS and PBS belong to the category of phosphate‑buffered solutions; based on typical formulations, their constituent components are largely identical, with only minor differences in concentration. In this sense, DPBS can be regarded as a specific type of PBS.
