Analysis of the causes for failure to stain with FITC and corresponding solutions?

1. The cell aggregates have not been dissociated. Solution: We recommend setting the centrifugal force at 500–1000 × g. If the centrifugal force is too low, only intact cells will be collected, while apoptotic cells and debris will be lost, resulting in an underestimation of the apoptosis rate. 2. A single‑color control was not performed. Solution: Fluorescence values and background signals vary between kits, and the gate positions differ as well. For example, compared to Li Ji, domestically produced Me* tends to shift left, while domestically produced Yi* shifts right. Therefore, we advise customers to set up single‑color gates for accurate analysis. 3. There are too many cells. Solution: If no signal is observed in the image, it indicates that the signal is beyond the detection range. We recommend adjusting the parameter settings to analyze approximately 10,000 cells. 4. FITC staining is unsuccessful. Solution: Freeze‑thaw cycles can adversely affect FITC labeling; therefore, we recommend avoiding cryopreservation for products from Li Ji. 5. Dye concentration, incubation time, and other experimental conditions are also key factors influencing staining. Solution: Different cell types may require distinct optimal conditions, which should be carefully determined through experimentation. 6. The intrinsic apoptotic stimulus provided to the cells is insufficient. Solution: Adjust the duration and concentration of the drug treatment. 7. Cell condition—e.g., cells undergoing excessive proliferation may exhibit reduced sensitivity to apoptotic stimuli. Solution: Verify and confirm the cells’ physiological state. 8. For adherent cells, prolonged digestion can damage the phosphatidylserine (PS) on the cell membrane, reducing the number of available Annexin V binding sites. Solution: Optimize cell digestion time and avoid over‑digestion.
