Is the CCK‑8 assay solution toxic to cells?
CCK‑8 solution itself exhibits slight toxicity due to the high concentration of PMS. However, when CCK‑8 is added to the culture medium, it becomes non‑toxic because it is diluted tenfold. Consequently, prolonged incubation—such as overnight or for several days—is feasible. Moreover, the same cell culture can be used for other proliferation assays after CCK‑8 treatment, including crystal violet staining, neutral red uptake, or DNA fluorescence assays. Since different cell types vary in their tolerance to CCK‑8, it is advisable to first assess cell viability following CCK‑8 addition before undertaking extended incubation periods.
