What is the price of the BCA protein quantitation kit? (How to use the protein quantitation kit)

The BCA (bicinchoninic acid) assay is currently one of the most widely used methods for determining protein concentration. It is based on the biuret reaction: under alkaline conditions, proteins reduce Cu²⁺ to Cu⁺, which then forms a purple-colored complex with the BCA reagent, exhibiting a strong absorbance at 562 nm. The amount of this reaction product is directly proportional to the protein concentration. By measuring the absorbance at 562 nm and comparing it to a standard curve, the concentration of the protein sample can be calculated. This method is rapid, sensitive, stable, and reliable, with a very low coefficient of variation across different protein types. The kit includes a stable protein standard solution for constructing the standard curve.

The BCA protein assay kit can be used for both tube‑based and microplate‑based assays. The tube method requires a relatively large protein sample (100 μL) and working reagent (2 mL), with a protein-to‑working‑reagent ratio of 1:20 (v/v). The measurable protein concentration range is 20–2000 μg/mL; with the enhanced protocol, detection down to 5 μg/mL is possible. The microplate method is simple and convenient, requiring only a small volume of protein sample (10–25 μL) and working reagent (200 μL), at a protein-to‑working‑reagent ratio of either 1:20 (v/v) or 1:8 (v/v). The measurable protein concentration range is 20–2000 μg/mL, and the enhanced protocol allows detection down to 5 μg/mL.

BCA Protein Quantitation Kit Instructions for Use

I. Preparation of Standard Solutions and Working Solutions

1. Prepare the BSA standard curve (linear range: 20–2000 μg/mL, or 5–250 μg/mL). Two preparation methods are provided for each concentration range; select the appropriate method based on your preference. [Note]: The BSA standard solution has a concentration of 2 mg/mL, and the diluent is the same as that used to dissolve the protein samples. In principle, the standard should be diluted in the same solvent as the sample; however, water or 1× PBS may also be used for dilution.

BSA Standard Preparation Protocol (After preparation, the solution can be stored at 4°C and reused multiple times)

2. Prepare the BCA working solution

(1) Calculate the total volume of BCA working solution required. Total BCA working solution volume = (number of standards + number of samples to be tested) × number of replicates × the volume of BCA working solution needed per sample. [Note]: For the tube‑based assay, add 2.0 mL of BCA working solution to each sample; for the microplate assay, add 200 μL of BCA working solution to each sample.

(2) Prepare the BCA working solution: Add 1 volume of BCA Reagent B to 50 volumes of BCA Reagent A (A:B = 50:1), and mix thoroughly. [Note]: Store the BCA working solution in a sealed container; it remains stable at room temperature for 24 hours.

II. Testing Methods

1. Test tube assay method (sample: BCA working solution = 1:20)

(1) Add 100 μL of the standard solution and 100 μL of the sample to be tested into the reaction tubes.

(2) Add to each tube
