How to deal with black mold contamination (black mold remover)

Whether the black glue worm is a living organism or a non‑living entity remains a subject of debate in the scientific community; for now, it is provisionally regarded as a microorganism. Often originating from serum, it is a common cellular contaminant that lives in symbiosis with cells, propagating through successive cell passages and typically remaining unaffected by antibiotics. The black glue worm competes with cells for nutrients and resources, adversely affecting cell growth and, in severe cases, leading to cell death. At present, many cell lines are contaminated with this organism, significantly compromising cell culture and downstream experimental outcomes.

Characteristics of the black glue worm

• The culture medium is no different from the standard culture medium;

• Under low magnification, they appear as black dots and are indistinguishable from cellular debris.

• Under high magnification, punctate and lamellar motile bodies can be observed, exhibiting Brownian motion that mimics cellular debris and cellular secretions.

• It is inversely proportional to the number of cells: the more cells there are, the less it is; the fewer cells there are, the more it increases. Replacing the culture medium or washing the cells does not remove it.

• Cells grow slowly and consume nutrients rapidly, requiring frequent medium changes; the longer they are cultured, the more their condition deteriorates.

Solution

• If the cells are in acceptable condition, the quickest and most effective approach is to use a black‑fly‑removal agent to eliminate black flies, while ensuring that the serum in the culture system is high‑quality serum free of black fly and protozoan contamination. Subsequently, increase the cell density and continue culturing for 2–3 passages to achieve clearance.

• If the cells are in poor condition, it is recommended to discard them after disinfection.

• Cell culture media contaminated with black mold do not become turbid, and their color and transparency remain unchanged. The contamination typically resolves spontaneously as cell proliferation intensifies. No special treatment is required beyond replacing the serum; increasing the seeding density can help improve cell viability. At present, the only effective preventive measure against black mold contamination is to use high‑quality serum.
