Cryopreservation method 1: Place the cryovial in a 4°C incubator for 30–60 minutes → transfer to −20°C for 30 minutes* → then store at −80°C for 16–18 hours (or overnight) → finally, transfer to the vapor phase of a liquid nitrogen tank for long-term storage.
Cryopreservation method 2: Place the cryovial in a programmable freezer set to a controlled cooling rate of 1–3°C per minute until the temperature reaches below −80°C, then transfer it to the vapor phase of a liquid nitrogen tank for long-term storage. *Note: Do not leave the vial at −20°C for more than 1 hour to prevent the formation of excessively large ice crystals, which can lead to significant cell death. Alternatively, you may skip this step and directly place the vial in a −80°C freezer; however, this will result in a lower survival rate.
