Product Description
All-in-One RT MasterMix (with dsDNase) is a highly efficient, convenient, and high-quality kit for single-stranded cDNA synthesis. It contains all the components required for reverse transcription, including a thermostable M-MLV GIII reverse transcriptase with its reaction buffer, RNase inhibitor, dNTPs, Oligo(dT)20VN, and random primers. To initiate the reaction, simply add your RNA template and water. The resulting cDNA is primarily used for subsequent qPCR analysis.
Extracted RNA samples often contain genomic DNA (gDNA) contamination, which can lead to the concurrent amplification of gDNA and cDNA in qPCR, compromising quantitative accuracy. This kit efficiently removes gDNA using dsDNase, which specifically degrades the DNA strand in double-stranded DNA or DNA–RNA hybrids and is rapidly and irreversibly inactivated at reverse transcription temperatures. Compared with traditionally used DNase I, it eliminates the need for additional EDTA treatment, streamlining the experimental workflow and reducing the risk of inhibiting the reverse transcription reaction.
This kit allows both reverse transcription and genomic DNA (gDNA) removal to be performed in a single tube, offering simple operation and effectively reducing the risk of sample contamination and RNA degradation associated with complex pipetting procedures. However, if genomic contamination is severe, it is recommended to use a protocol that separates gDNA removal from reverse transcription.
