Product Description
The Annexin V-FITC/PI Apoptosis Assay Kit is a reagent kit that combines Annexin V and PI, designed to detect the early stages of apoptosis and to distinguish between early apoptotic cells and late apoptotic or necrotic cells.
Annexin V is a member of the intracellular annexin protein family and selectively binds to phosphatidylserine (PS) in a calcium‑dependent manner. Under normal conditions, PS is localized on the inner leaflet of the cell membrane. The externalization of PS to the outer leaflet is a hallmark of early apoptosis in various cell types. This critical event can be detected using Annexin V labeled with FITC—Annexin V‑FITC—via flow cytometry or fluorescence microscopy. FITC emits yellow‑green fluorescence.
PI (propidium iodide) is a nuclear staining reagent that can label DNA and emits red fluorescence upon intercalation into DNA. PI cannot penetrate intact cell membranes, but it can enter necrotic cells or cells in the late stages of apoptosis that have lost membrane integrity.
When Annexin V-FITC and PI are used in combination, PI is excluded from live cells (Annexin V-/PI-) and early apoptotic cells (Annexin V+/PI-), whereas late apoptotic and necrotic cells are simultaneously stained by both FITC and PI, resulting in double positivity (Annexin V+/PI+).
The FITC exhibits maximum absorption and excitation wavelengths of 494 nm and 518 nm, respectively, while the PI–DNA complex shows peak absorption and emission wavelengths of 535 nm and 617 nm, respectively.
