Product Description
The molecular formula of CFDA-SE is C 29 H 19 NO 11 Its molecular weight is 557.47, and its CAS number is 150347-59-4. The English name of CFDA‑SE is Carboxyfluorescein diacetate, succinimidyl ester. CFDA‑SE can be used for cell tracking as well as for assessing cell proliferation. It can pass through an intact cell membrane; once inside the cell, it is hydrolyzed by intracellular esterases into CFSE. CFSE then undergoes random, irreversible covalent binding with lysine residues or other amino groups on intracellular proteins, thereby labeling these proteins. After incubation with the fluorescent probe CFDA‑SE for approximately 24 hours, cells are fully labeled. The fluorescence in non‑dividing cells labeled with CFDA‑SE is highly stable, with labeling lasting for several months.
CFDA‑SE–labeled cells exhibit green fluorescence; for detection, an excitation wavelength of 488 nm and an emission wavelength of 518 nm can be used, with the FL1 detection channel on a flow cytometer. In addition to assessing cell proliferation by flow cytometry, live cell counts can also be quantified using a fluorescent microplate assay, or uniform staining can be visualized and tracked under a fluorescence microscope.
CFDA‑SE–labeled dividing cells exhibit a fluorescence intensity that decreases by half with each division. Consequently, flow cytometry can distinguish between undivided cells and those that have undergone one division—whose fluorescence is reduced to 1/2—those that have divided twice—reduced to 1/4—and so on. Thus, the fluorescence intensity diminishes sequentially according to the pattern 1/2, 1/4, 1/8, 1/16, and so forth. Using CFDA‑SE, flow cytometric analysis yields distinct peaks, with each peak corresponding to a specific number of cell divisions; moving from right to left, these peaks typically represent cells that have undergone 0, 1, 2, 3, and higher divisions. As the number of divisions increases, cells that have not divided or have undergone only a few divisions gradually become less abundant and may eventually be undetectable. CFDA‑SE can reliably track up to eight or even more rounds of division. Cells labeled with CFDA‑SE are suitable for both in vitro and in vivo proliferation studies and do not stain neighboring cells. CFDA‑SE is most commonly employed for assessing lymphocyte proliferation, but it can also be used to study the proliferation of fibroblasts, natural killer cells, hematopoietic progenitor cells, and other cell types. When combined with other fluorescent probes, CFDA‑SE enables the acquisition of additional information about cells at different stages of division. Our CFDA‑SE probe is supplied as a 25 mg powder formulation.
