Product Description
Hoechst 33258 is a blue fluorescent dye that can penetrate cell membranes, exhibits low cytotoxicity, and emits intense blue fluorescence upon intercalation into double-stranded DNA. It is a DNA‑specific stain that binds preferentially to A–T base pairs; it stains dead cells or cells fixed with 70% cold ethanol immediately, whereas staining of live cells occurs progressively, reaching saturation within 10 minutes. Under a fluorescence microscope, the nuclei of live cells display diffuse, uniform fluorescence; during apoptosis, densely stained, compact granular fluorescent aggregates become visible in the nucleus or cytoplasm. Hoechst 33258 is commonly used for detecting apoptosis, with stained samples examined by fluorescence microscopy or analyzed by flow cytometry, and it can also be employed for routine visualization of cell nuclei and mitochondria.
Hoechst is often used as a substitute for other nucleic acid dyes such as DAPI. The key difference between the two is that, compared with DAPI, Hoechst 33258 exhibits greater lipophilicity, allowing it to penetrate intact cell membranes more effectively. For nuclear staining, the recommended working concentration of Hoechst 33258 is 0.5–10 μg/mL.
