Product Description
Hoechst 33342 is a blue fluorescent dye that can penetrate cell membranes, exhibits low cytotoxicity, and emits intense blue fluorescence upon intercalation into double-stranded DNA. Due to their ability to bind to DNA, Hoechst‑series dyes are also frequently used for visualizing mitochondrial DNA.
Hoechst 33258 and Hoechst 33342 are two closely related Hoechst dyes; both are excited at 350 nm in the ultraviolet range and emit at 461 nm. These dyes are often used as alternatives to the fluorescent dye DAPI. Hoechst 33342 exhibits greater lipophilicity than 33258, allowing it to penetrate intact cell membranes more effectively; in some experiments, Hoechst 33258 shows markedly reduced cellular permeability. For nuclear staining, the recommended final working concentration of Hoechst 33342 is 0.5–5 μM; in sensitive assays, it is advisable to test multiple staining concentrations to optimize the staining outcome.
