Product Description
When cells undergo apoptosis, certain DNA endonucleases are activated. These enzymes cleave genomic DNA between nucleosomes, generating DNA fragments of 180–200 base pairs, which appear as a characteristic ladder‑like pattern on agarose gel electrophoresis. When genomic DNA suffers double‑strand or single‑strand breaks, numerous sticky 3′‑OH termini are produced. Under the catalytic action of terminal deoxynucleotidyl transferase (TdT), these ends can be labeled with dUTP, enabling direct detection of apoptotic cells by fluorescence microscopy or flow cytometry. This approach is known as terminal deoxynucleotidyl transferase‑mediated nick‑end labeling (TUNEL). Because normal or proliferating cells exhibit virtually no DNA fragmentation and thus lack 3′‑OH groups, they are rarely stained. The TUNEL assay allows in situ staining of intact individual apoptotic nuclei or apoptotic bodies, accurately reflecting the hallmark biochemical and morphological features of apoptosis, and can detect even very low numbers of apoptotic cells, making it widely used in apoptosis research.
This kit has a broad range of applications and can be used to detect apoptosis in frozen or paraffin‑embedded tissue sections, as well as in cultured adherent or suspension cells. It selectively identifies apoptotic cells, while excluding necrotic cells and cells exhibiting DNA strand breaks induced by radiation or pharmacological treatment. The assay is rapid, requiring only a single staining step; after washing, detection can be performed immediately.
