Product Description
The live-cell apoptosis assay kit is based on fluorescent caspase inhibitors. These inhibitors are cell-permeable and non‑cytotoxic. Once inside the cell, the caspase inhibitors covalently bind to active caspases. Caspase‑1 plays a key role in the activation of pro‑inflammatory cytokines and in the apoptotic process. Caspase‑1 has been shown to exhibit substrate specificity for the peptide sequence Tyr‑Val‑Ala‑Asp (YVAD). This kit employs FAM‑YVAD‑FMK as a fluorescent probe for detecting caspase‑1 activity. In apoptotic cells, FAM‑YVAD‑FMK irreversibly binds to activated caspase‑1. Upon binding to caspase‑1, the fluorescent probe remains within the cell. Since this inhibitor selectively targets caspase‑1 without halting apoptosis, a variety of parameters can be used to monitor the apoptotic process.
This kit is designed to detect apoptosis by measuring caspase‑1 activation in living cells. It is intended for the quantitative assessment of activated caspase‑1 activity in apoptotic cells or for screening caspase‑1 inhibitors. The green fluorescent probe FAM‑YVAD‑FMK enables direct detection of activated caspase‑1 in apoptotic cells using fluorescence microscopy, flow cytometry, or a fluorescence microplate reader. The kit provides an optimized assay protocol that includes all necessary components.
