The Caspase 3 & 7 Activity Cell Apoptosis Assay Kit is a family of tools designed to assess cellular functions, including cell viability, cytotoxicity, apoptosis, mitochondrial membrane potential, and the cell cycle. Each assay offers detection options with distinct fluorescent colors. These highly efficient assays provide a powerful approach for investigating cellular functional activities from multiple perspectives. The Caspase 3/7 Activity Cell Apoptosis Assay Kit detects apoptosis by measuring the activity of caspase‑3/7. Since caspase‑3 (CPP32/apopain) plays a critical role in the initiation of apoptosis, it has been widely recognized as a reliable marker of apoptotic events. Caspase‑3 possesses a specific substrate peptide recognition sequence—Aspartate‑Glutamate‑Valine‑Aspartate (DEVD). This kit employs Z‑DEVD‑Rh110‑DVED‑Z as a fluorescent probe for detecting caspase‑3 activity. Upon cleavage by caspase‑3, the Rh110 peptide is fragmented, releasing Rh110, which emits intense green fluorescence. Fluorescence intensity is measured at 520–530 nm, with an excitation wavelength of 340–350 nm.
This kit provides all essential components and an optimized assay protocol. The assay is robust and reliable, suitable for high-throughput analysis on a variety of fluorescence-based platforms, including microplate assays, immunohistochemistry, and flow cytometry. When using a 96-well plate, add 100 µL of reagent per well; the reagents supplied in this kit are sufficient for 200 assays. For a 384-well plate, use 25 µL of reagent per well; the reagents provided are sufficient for 800 assays.
