The Caspase 8 Activity Cell Apoptosis Assay Kit is used for detecting apoptosis by measuring Caspase 8 activity. Caspase 8 is a caspase protein encoded by the CASP8 gene. In neurodegenerative diseases, such as Huntington’s disease, Caspase 8 plays an important role. Caspase 8 has been shown to exhibit substrate specificity, with an Ile-Glu-Thr-Asp (IETD) peptide motif. This kit employs (Ac-IETD)2‑R110 as a fluorescent probe to detect Caspase 8 activity. Cleavage of the R110 peptide by Caspase 8 generates intense green fluorescence, with emission wavelengths between 520 and 530 nm and excitation wavelengths between 480 and 500 nm. This property makes the kit compatible with FITC filters.
This kit provides all necessary components and an optimized protocol. The assay delivers robust, rapid results and is well suited for high-throughput screening. It not only enables quantitative measurement of caspase‑8 activity in apoptotic cells but also facilitates the identification of caspase‑8 inhibitors.
