Product Description
A decrease in mitochondrial membrane potential (ΔΨm) is a key hallmark of early apoptosis. This decline in membrane potential arises from alterations in mitochondrial membrane permeability, primarily driven by pro-apoptotic proteins—such as the formation of Bax dimers and the activation of Bid, Bak, and Bad—which induce the opening of pores in the outer mitochondrial membrane. Concurrent with the activation of these proteins, cytochrome c is released from the mitochondria into the cytoplasm.
JC‑1 is an ideal fluorescent probe widely used for detecting ΔΨm, suitable for cells, tissues, or purified mitochondrial samples. Its detection principle relies on a membrane potential–dependent fluorescence state transition: when the mitochondrial membrane potential is high, JC‑1 aggregates in the mitochondrial matrix to form polymers, emitting red fluorescence; when the mitochondrial membrane potential is low, JC‑1 fails to aggregate in the matrix and exists as monomers, yielding green fluorescence. The shift from red to green fluorescence upon JC‑1 aggregation provides a straightforward readout of membrane potential decline and can also serve as an early biomarker of apoptosis.
The JC‑1 monomer has a maximum excitation wavelength of 510 nm and a maximum emission wavelength of 527 nm; the JC‑1 aggregate exhibits a maximum excitation wavelength of 585 nm and a maximum emission wavelength of 590 nm. This kit is easy and rapid to use, and can be detected using a flow cytometer, a fluorescence microscope, or a fluorescence microplate reader.
