Product Description
A hallmark of apoptosis is the degradation of cellular chromosomal DNA, a phenomenon that is quite widespread. This degradation is highly specific and orderly, yielding DNA fragments of lengths that are integer multiples of approximately 180–200 base pairs, which manifest as a characteristic ladder‑like pattern on agarose gel electrophoresis. This kit employs the TUNEL assay, in which terminal deoxynucleotidyl transferase (TdT) catalyzes the incorporation of biotin‑labeled dUTP (Biotin‑X‑dUTP) at the 3′‑OH ends of fragmented DNA in apoptotic cells. The incorporated biotinylated nucleotides then specifically bind to streptavidin conjugated with horseradish peroxidase (Streptavidin‑HRP). Finally, under HRP catalysis, DAB staining visualizes apoptotic cells, allowing their detection and enumeration by conventional light microscopy. Because normal or proliferating cells exhibit minimal DNA fragmentation and thus lack 3′‑OH groups, they are rarely stained. The TUNEL assay enables selective, direct in situ detection of apoptotic cells, distinguishing them from necrotic cells or those undergoing DNA strand breaks induced by irradiation or pharmacological treatments, making it a faster and more straightforward method for assessing apoptosis.
