Trypsin, often referred to as pancreatic enzyme, is a serine protease that cleaves the carboxyl‑terminal bonds of lysine and arginine residues in polypeptide chains. During the isolation and culture of tissue cells, as well as in in vitro cell culture, trypsin digestion solutions are routinely used to hydrolyze intercellular proteins, thereby dissociating tissues or cell aggregates into single cells.

Pancreatic enzymes exhibit their highest digestive activity at 37°C and pH 8.0. A commonly used working concentration of pancreatic enzyme is 0.25%. EDTA chelates calcium and magnesium ions; when added to a pancreatic enzyme solution, it accelerates the hydrolysis of intercellular junction proteins, thereby enhancing digestion. Phenol red is a pH indicator: it appears purplish‑red in alkaline solutions, orange‑red in acidic solutions, and pink near neutral pH.

This product contains 0.25% trypsin, is dissolved in Hank’s buffer, supplemented with 0.9 mM EDTA, and includes a phenol red indicator; it has been sterilized by filtration through a 0.22 μm membrane.
