Product Description
The Cell Cycle and Apoptosis Analysis Kit is a reagent kit that employs the classic propidium iodide staining (PI staining) method for analyzing cell cycle progression and apoptosis.
Propidium iodide (PI) is a fluorescent dye that binds to double-stranded DNA. Upon binding to double-stranded DNA, PI emits fluorescence, with the fluorescence intensity directly proportional to the amount of double-stranded DNA present. After staining intracellular DNA with propidium iodide, flow cytometry can be used to quantify DNA content; based on the distribution of DNA content, cell cycle and apoptosis analyses can then be performed. After propidium iodide staining, if the fluorescence intensity of G0/G1-phase cells is set to 1, then the theoretical fluorescence intensity of G2/M-phase cells, which contain a doubled genome, is 2, while S-phase cells undergoing DNA replication exhibit fluorescence intensities between 1 and 2. Apoptotic cells show markedly reduced staining—i.e., a fluorescence intensity below 1—due to nuclear condensation and DNA fragmentation, resulting in the loss of portions of genomic DNA during staining. Consequently, on flow cytometry fluorescence plots, apoptotic cells give rise to the so‑called sub‑G1 peak. During apoptosis, cytoplasmic and chromatin condensation, along with nuclear fragmentation, give rise to apoptotic bodies, thereby altering the cell’s light-scattering properties. In the early stages of apoptosis, the cell’s forward-angle light scatter decreases markedly, while its side‑angle light scatter either increases or remains unchanged. In the late stages of apoptosis, both forward- and side‑angle light scatter are reduced. Consequently, changes in cellular light scattering can be monitored using flow cytometry to assess apoptotic events.
This kit is typically used for analyzing the cell cycle and apoptosis in cultured adherent or suspension cells. If applied to tissue samples, the tissue must first be dissociated into a single-cell suspension before analysis can be performed.
