Wheat germ agglutinin, labeled with AF594 (hereinafter referred to as “AF594”) WGA ”), is a lectin that binds to N‑acetyl‑D‑glucosamine and sialic acid, and may be the brightest WGA conjugate among those exhibiting bright red fluorescence. It is currently one of the most extensively studied and widely applied classes of lectins. Because WGA binds to glycoconjugates, its derivatives and conjugates are extensively used for labeling cell membranes and fibrotic scar tissues in fluorescent imaging and analysis. The sugar‑binding specificity of WGA is linked to the sequence of β‑1,4‑GlcNAc‑linked residues, which is associated with chitinase activity. Each monomer contains two identical, non‑interacting binding sites that are complementary to three or four β‑1,4‑GlcNAc units. Among the monosaccharides examined, only GlcNAc binds to WGA; mannose does not bind, while GalNAc exhibits only weak binding. WGA displays high‑affinity binding to internal GlcNAc residues within large oligosaccharides containing repeating sequences of Galβ(1–4)GlcNAcβ(1–3), i.e., poly‑lactosamine‑type glycans. N‑acetylneuraminic acid participates only in low‑affinity interactions with WGA. WGA demonstrates a complex pattern of carbohydrate specificity, making it useful for structural analysis of intricate carbohydrates.

    •

Q: When using AF594 WGA for fluorescent labeling of cell membranes, which sample types are suitable?

A: It can be used with live or fixed cells and is widely employed for labeling cell membranes and fibrotic scar tissue in fluorescence imaging and analysis. Wheat germ agglutinin can also be applied to paraffin‑embedded sections; for paraffin section staining, permeabilization is not recommended, as it may cause the dye to enter cellular organelles and result in non‑specific staining.
