  • Product Description

Proteinase K, with a molecular mass of approximately 29.3 kDa, cleaves peptide bonds at the carboxyl terminus of both aliphatic and aromatic amino acids, making it a serine protease with broad substrate specificity. It remains active over a wide pH range (pH 4–12.5). Its typical working concentration is 50–100 µg/mL; however, its activity can be enhanced in the presence of denaturants such as 1% SDS. Therefore, the appropriate working concentration should be determined based on factors such as the buffer composition (e.g., presence of SDS or urea), pH, and temperature. At commonly used concentrations, EDTA, Triton X‑100, Tween 20, Sarkosyl, and guanidine hydrochloride have little effect on Proteinase K activity.

It is commonly used in experiments such as genomic DNA extraction and the preparation of chromosomal DNA for pulsed-field gel electrophoresis.

    Transportation and Storage

Blue ice transport. Store at −20°C; shelf life: 12 months. [Note]: Avoid repeated freeze–thaw cycles.

    Instructions for Use

    1. Preparation of Proteinase K solution: Typically, a 20 mg/mL Proteinase K solution is prepared. For example, dissolve 200 mg of Proteinase K in 9.5 mL of ddH₂O, gently shaking until completely dissolved. Avoid vortexing; then add water to bring the final volume to 10 mL, and aliquot into small portions for storage at −20°C.

    2. Adjust the final concentration to 50–100 µg/mL, then pipette an appropriate volume of Proteinase K solution into the sample to be digested. Incubate at 55–65°C; the optimal incubation temperature is 58°C. Incubation time ranges from 15 minutes to 48 hours, depending on the characteristics of the sample.

    Precautions

    1. This product is intended solely for scientific research and must not be used for clinical diagnosis, treatment, or any other purposes.

    2. If you need to further dilute 20 mg/mL proteinase K, it is recommended to use 10 mM Tris‑HCl buffer at pH 7.4–7.8 for the dilution.

    3. This product is hazardous to humans. During handling, be sure to wear a lab coat, disposable gloves, and a mask to avoid direct skin contact or inhalation.
