Product Description
Proteinase K, with a molecular mass of approximately 29.3 kDa, cleaves peptide bonds at the carboxyl terminus of both aliphatic and aromatic amino acids, making it a serine protease with broad substrate specificity. It remains active over a wide pH range (pH 4–12.5). The typical working concentration is 50–100 µg/mL; however, its activity can be enhanced in the presence of denaturants such as 1% SDS. Therefore, the optimal working concentration should be determined based on factors such as the buffer composition (e.g., presence of SDS or urea), pH, and temperature. Commonly used concentrations of EDTA, Triton X‑100, Tween 20, Sarkosyl, and guanidine hydrochloride have little effect on Proteinase K activity.
It is commonly used in experiments such as genomic DNA extraction and the preparation of chromosomal DNA for pulsed-field gel electrophoresis.
