Blasticidin S is a nucleoside antibiotic derived from the bacterium Streptomyces griseochromogenes, known in Chinese as Mie-wen-su S, Sha-dao-wen-jun-su S, or Dao-wen-san; it is typically supplied as its hydrochloride salt. Blasticidin S is commonly used to select mammalian cell lines stably transfected with plasmids carrying the bsr, BSD, or bls gene (often designated as bsrr, bsdr, or Blastr), exhibiting a rapid and potent mode of action: even at very low antibiotic concentrations, it can swiftly kill cells lacking the resistance gene.

This product is formulated at a concentration of 10 mg/mL in HEPES‑buffered solution (pH 7.4), sterilized by filtration through a 0.22 µm membrane, and can be used directly for cell culture. The 10 mg and 50 mg packages are supplied as a powder.

Blasticidin S specifically inhibits protein synthesis in prokaryotic or eukaryotic cells by interfering with ribosome‑binding peptides. Currently, three blasticidin‑S resistance genes have been identified: the acetyltransferase gene bls, isolated from Streptoverticillum sp.; and two deaminase genes, bsr, derived from Bacillus cereus, and BSD, isolated from Aspergillus terreus. The bsr and BSD genes are the most commonly used selection markers for generating stable cell lines in mammalian and plant cells. Blasticidin S can also be employed for screening prokaryotic cells such as Escherichia coli.

Blasticidin S exhibits a rapid and potent mode of action, inducing swift cell death even at very low antibiotic concentrations. Escherichia coli is typically sensitive to concentrations of 50 μg/mL, whereas mammalian cells are sensitive to much lower levels, ranging from 2 to 10 µg/mL. Cell death occurs rapidly, and stable mammalian cell lines resistant to Blasticidin S can usually be established in less than one week.
