Product Description
Puromycin is an amino‑nucleoside antibiotic derived from Streptomyces alboniger; its Chinese name is transliterated as “Piaoling Meisu”. It is commonly used to select eukaryotic or prokaryotic polyclonal or monoclonal cells expressing the pac (puroR) gene introduced by plasmid transfection/transformation, viral infection, or other methods. Puromycin is used both to select stable cell lines and to maintain them. The 10 mg/ml formulation of this product is filter‑sterilized and can be used directly in cell culture.
Puromycin is characterized by its rapid action on cells, typically killing 99% of cells that do not express the pac gene within 2 days.
In Gram-positive bacteria, animal cells, or insect cells, puromycin inhibits or kills cells by blocking protein synthesis. Its mechanism of action involves puromycin, an analog of the 3′‑terminal end of an aminoacyl‑tRNA molecule, which can bind to the A site of the ribosome and become incorporated into the growing polypeptide chain. Once bound to the A site, puromycin does not participate in subsequent reactions, leading to premature termination of translation and the release of an immature polypeptide bearing puromycin at its C‑terminus.
The pac gene encodes puromycin N-acetyltransferase, a gene originally identified in Streptomyces alboniger. When expressed, it confers resistance to puromycin—a property that is widely used today to select stable mammalian cell lines expressing the pac gene. For example, many commercially available lentiviral vectors carry the pac gene (often labeled as “puror” on the plasmid map), enabling the selection of cells stably expressing a specific gene with puromycin. Puromycin can also be employed to screen bacterial strains, such as E. coli, and yeast strains that express the pac gene.
