Product Description
Zeocin Namely phleomycin D1, known in Chinese as “bo lai mei su” or “fu cao mei su D1,” is a member of the bleomycin/phleomycin antibiotic family, isolated from a mutant strain of Streptomyces verticillus. It exhibits inhibitory activity and cytotoxicity against bacteria, fungi (including yeasts), plants, and mammalian cells.
Zeocin It is an alkaline, water-soluble, copper‑ion‑chelating glycopeptide antibiotic. A Cu²⁺‑chelated Zeocin solution appears blue and is inactive. Upon entering the cell, the Cu²⁺ on Zeocin is reduced to a monovalent copper ion (Cu⁺) and subsequently scavenged by intracellular sulfhydryl compounds, thereby activating Zeocin. The activated compound can then bind to cellular DNA and induce DNA strand breaks, ultimately leading to cell death.
The protein conferring resistance to Zeocin is encoded by the Shble gene from Streptoalloteichus hindustanus; it produces a 14 kDa protein that binds Zeocin at a specific ratio, preventing its incorporation into cellular DNA and inhibiting its DNA‑cleaving activity, thereby rendering cells resistant to Zeocin. Consequently, Zeocin can be used to select polyclonal or monoclonal cell lines expressing the Zeocin‑resistance gene, or for the maintenance culture of such cell lines.
Zeocin It is effective for the vast majority of aerobic cells and is commonly used for screening bacteria (such as Escherichia coli), eukaryotic microorganisms (such as yeast), and animal and plant cells. For E. coli, a recommended concentration of 25–50 μg/mL is advised (using low-salt LB medium, with NaCl not exceeding 5 g/L); for yeast, 50–300 μg/mL is recommended (in YPD or a minimal medium); and for mammalian cells, 50–1000 μg/mL is suggested (in an appropriate culture medium, which may vary depending on the cell line). In practical applications, it is essential to test a series of Zeocin concentrations for each specific cell line to determine the optimal working concentration.
