Product Description
Evagreen is a DNA-binding dye used in real-time quantitative PCR (qPCR). Its numerous advantages make it far superior to SYBR Green I. In addition to exhibiting similar spectral properties, Evagreen possesses three key features that distinguish it from SYBR Green I:
Evagreen exhibits far less inhibition of PCR than SYBR Green I, allowing qPCR assays to be performed using rapid PCR cycling conditions. Moreover, Evagreen can be used at higher concentrations, yielding amplification signals that are significantly stronger than those obtained with SYBR Green I. The higher concentration of Evagreen also eliminates the “dye redistribution” artifact, making it suitable for both multiplex PCR and high-resolution melting (HRM) analysis. This analytical approach is increasingly being employed for post‑PCR genotyping and heteroduplex analysis. In contrast, because SYBR Green I exerts substantial inhibitory effects on PCR, it must be used at very low concentrations; consequently, it cannot address the dye redistribution issue arising from such low concentrations, precluding its use in multiplex PCR or HRM. Furthermore, dye redistribution can compromise the reliability of conventional melting curves, as DNA strands with lower melting temperatures may remain undetected under these conditions.
Evagreen exhibits exceptional stability and remains intact under standard storage, handling, and PCR conditions. The dye in buffer can be safely stored at room temperature or in the refrigerator and withstands multiple freeze–thaw cycles. In contrast, SYBR Green I is unstable and, upon degradation, exerts a stronger inhibitory effect on PCR.
Evagreen reduces cell membrane permeability, making it safer than SYBR Green 1. Independent laboratory tests have shown that Evagreen exhibits neither mutagenicity nor cytotoxicity. In contrast, although SYBR Green I itself has very low mutagenic potential, it may inhibit the cell’s normal DNA repair mechanisms, thereby exerting a mutagenic‑enhancing effect. Given the widespread use of PCR, its safety merits careful consideration.
