Product Description
PicoGreen is an extremely sensitive fluorescent dye for double-stranded DNA that emits fluorescence only upon binding to dsDNA, with fluorescence intensity directly proportional to DNA concentration. In the presence of ssDNA, RNA, and monomeric nucleotides, it can selectively detect dsDNA at concentrations as low as 25 pg/mL. Compared with conventional DNA quantitation methods, PicoGreen offers advantages such as high sensitivity, strong specificity, excellent tolerance to interfering substances, a broad linear range, and ease of use, making it well suited for precise determination of DNA concentrations in applications like cDNA library construction, DNA subcloning, and PCR product analysis. Compared with traditional approaches, the PicoGreen dsDNA Quantitation Reagent provides the following benefits:
Sensitivity: It is more sensitive than UV absorbance readings and can help conserve samples.
High specificity: It binds exclusively to dsDNA, exhibiting high specificity and remaining unaffected by common sample contaminants. In contrast, conventional UV absorbance methods are susceptible to interference from single-stranded DNA, proteins, RNAs, and other substances present in the sample.
Good tolerability: It tolerates high concentrations of salt, urea, ethanol, chloroform, detergents, proteins, or agarose, allowing direct quantification of PCR products without prior DNA purification from the reaction mixture.
Wide linear range: PicoGreen dsDNA dye quantifies DNA concentration with good linearity over the range of 100 pg/mL to 100 ng/mL; it is compatible with fluorometers.
Easy to use: Add a dye to the sample, allow 5–6 minutes for staining, and then take a reading; this method is compatible with both 96‑well and 384‑well microplates. It is applicable to a wide range of assays, including fluorescence‑based enzyme‑linked immunosorbent assays, DNA‑sample analysis, PCR analysis, chip‑based assays, DNA‑damage assessment, enzyme‑activity measurements, quantitative genomic DNA analysis, as well as the detection of double‑stranded DNA in complex mixtures and the quantification of viral DNA.
The assay exhibits linearity across four orders of magnitude and virtually no sequence dependence, enabling precise quantification of DNA from multiple sources, including genomic DNA, viral DNA, low‑input DNA, or PCR amplification products. Owing to its robustness and high sensitivity, this method has been incorporated into the Chinese Pharmacopoeia.
