Product Description
After cells or tissues are fixed with paraformaldehyde, formaldehyde, or other aldehyde‑based reagents, intracellular antigens can form aldehyde‑linked and carboxymethyl‑linked cross‑links that mask certain antigenic epitopes. Simultaneously, protein–protein cross‑linking further conceals these epitopes, leading to reduced staining signals in immunostaining and even false‑negative results. Therefore, prior to performing immunohistochemical staining, it is essential to carry out antigen retrieval or unmasking—breaking the cross‑links formed during fixation and restoring the antigen’s native three‑dimensional conformation. This enhances antigen detection, minimizes background staining, and improves assay accuracy.
Sodium citrate antigen retrieval solution (50×) is a widely used antigen retrieval buffer that can be employed for antigen retrieval in paraffin‑embedded and frozen tissue sections fixed with formaldehyde, paraformaldehyde, or other aldehyde‑based fixatives. It effectively disrupts protein–protein cross‑links induced by aldehyde fixation, thereby fully exposing antigen epitopes in paraffin‑embedded samples and significantly enhancing the quality of immunostaining.
Typically, paraffin‑embedded sections require antigen retrieval, whereas frozen sections can be stained without it. Antigen retrieval can enhance the immunostaining performance of paraffin sections and, to varying degrees, improve staining in frozen sections as well. When the immunostaining results from frozen sections are suboptimal, antigen retrieval should be considered.
Based on a requirement of 10 mL of antigen retrieval solution (1×) per slide, 100 mL of antigen retrieval solution (50×) can be used for antigen retrieval on 500 samples.
