Product Description
Firefly luciferase is a protein with a molecular mass of approximately 61 kDa that, in the presence of ATP, magnesium ions, and oxygen, catalyzes the oxidation of luciferin to oxyluciferin, concomitant with bioluminescence. Renilla luciferase is a protein with a molecular mass of about 36 kDa that, under aerobic conditions, catalyzes the oxidation of coelenterazine to coelenteramide, also accompanied by bioluminescence. Bioluminescence is a form of chemiluminescence, in which light energy is released through a chemical reaction, and it can be quantified using a luminometer or a liquid scintillation counter.

In DLR assays, the activities of Firefly luciferase and Renilla luciferase can be sequentially measured in a single sample. First, Firefly luciferase is assayed using luciferin as the substrate; subsequently, Renilla luciferase is measured with coelenterazine as the substrate. During the second assay, an inhibitor that blocks Firefly luciferase–catalyzed luminescence from luciferin is added, ensuring that only Renilla luciferase activity is detected. This enables dual‑luciferase reporter gene analysis. Leveraging the bioluminescent system of luciferase and its substrates, gene expression can be detected with high sensitivity and efficiency. Typically, transcriptional regulatory elements or the 5′ promoter region of the gene of interest are cloned upstream of the luciferase gene, while the 3′‑UTR is cloned downstream, generating a reporter‑gene plasmid. The plasmid is then transfected into cells, which are treated with appropriate agents before being lysed to measure luciferase activity. Changes in luciferase activity serve as an indicator of how the treatment modulates transcriptional regulation of the target gene. Renilla luciferase is more commonly used as an internal control for transfection efficiency, thereby accounting for variations in cell number and transfection efficiency.
Li Ji Bio’s Firefly & Renilla Dual-Luciferase Reporter Assay Kit features rapid detection, high sensitivity, a broad assay range, and no interference from endogenous cellular activity.
