Product Description
TBE is a commonly used nucleic acid electrophoresis buffer in biology, primarily employed for agarose gel electrophoresis and polyacrylamide gel electrophoresis of DNA. TBE offers strong buffering capacity, delivering superior resolution when separating fragments smaller than 1 kb and is well suited for extended electrophoresis runs. However, when used with agarose gels, TBE can promote high electroosmotic flow and, through its interaction with agarose to form non‑covalent tetrahydroxyborate complexes, reduce the recovery yield of DNA fragments; therefore, it is not recommended for DNA fragment recovery. When high resolution is not critical, either TAE or TBE may be used; for applications demanding higher resolution, lower‑concentration gels enhance the separation of high‑molecular‑weight nucleic acids, making TAE the preferred choice, whereas higher‑concentration gels improve the resolution of low‑molecular‑weight nucleic acids, favoring TBE.
The main components of TBE are Tris, boric acid, and EDTA. In 1× TBE buffer, the concentration of Tris is 89 mM, that of boric acid is 89 mM, and that of EDTA is 2 mM.
