Product Description
Cyanine 5 is widely used, not only for labeling biomolecules but also as a derivatization reagent for certain small molecules. Compared with other fluorescent reagents that label amino groups—such as FITC and TRITC—Cyanine 5 NHS ester exhibits superior reaction selectivity.
Cyanine5 NHS ester is a near-infrared dye that, in addition to labeling the amino groups of peptides, proteins, and oligonucleotides, is also widely used for in vivo imaging in small animals. By avoiding the visible-light region, where many biomolecules exhibit strong absorption, it absorbs near-infrared light, which penetrates biological tissues more deeply, while its emitted fluorescence is less affected by background autofluorescence in tissues, enabling signal generation even in deep tissues. In particular, within the 700–900 nm range, both water and hemoglobin exhibit minimal absorption, allowing near-infrared light to penetrate up to 15 cm into tissue. Moreover, these dyes offer a level of biosafety that cannot be achieved with UV‑absorbing dyes or isotopic labels. Cyanine 5 (Cy5) is the most commonly used near-infrared fluorescent dye.
Cyanine 5 NHS ester requires a cosolvent, but due to its lower hydrophobicity compared to Cyanine 7, it is more widely used. Li Ji Bio offers Cyanine 5 NHS ester in the form of triethylammonium salts, which are more soluble than the potassium‑salt forms offered by other companies. With comparable optical properties and reactivity, these triethylammonium salts provide superior solubility. NHS esters (succinimidyl esters) are commonly employed to label amino groups on biomolecules, while maleimides are frequently used to modify thiol groups.
Cyanine 5.5 NHS ester (succinimidyl ester) exhibits broad spectral properties similar to those of Alexa Fluor 680 NHS ester and DyLight 680 NHS ester, making it a suitable alternative.
