Product Description
iFluor dyes are a family of superior fluorescent labeling dyes that span the entire visible spectrum. All iFluor dyes exhibit excellent water solubility, and their hydrophilic nature minimizes the need for organic solvents. Moreover, iFluor dyes offer enhanced labeling performance compared to conventional fluorescent labels such as FITC, TRITC, Texas Red, Cy3, Cy5, and Cy7. Certain iFluor dyes outperform Alexa Fluor–based labels on specific antibodies. These dyes provide an exceptionally cost-effective alternative for labeling proteins and nucleic acids without compromising performance. Each iFluor dye has been specifically engineered to match the spectral characteristics of a corresponding Alexa Fluor or other well‑established label, such as DyLight dyes.
N‑succinimidyl esters have been shown to be excellent reagents for amine modification, as the resulting amide bonds are essentially identical to native peptide linkages and are highly stable. These reagents are generally stable and exhibit good reactivity and selectivity toward aliphatic amines. When using succinimidyl ester compounds in conjugation reactions, only a few factors need to be considered: 1. Solvent: In most cases, the reactive dye should be dissolved in anhydrous dimethylformamide (DMF) or dimethyl sulfoxide (DMSO). 2. Reaction pH: The labeling reaction between amines and succinimidyl esters is strongly pH‑dependent. Aminoreactive reagents react with deprotonated aliphatic amine groups, including terminal amines on proteins and the β‑amino group of lysine residues. Consequently, amine acylation reactions are typically carried out at pH values above 7.5. Protein modifications mediated by succinimidyl esters can usually be performed at pH 8.5–9.5. 3. Reaction buffer: When employing aminoreactive reagents, buffers containing free amines—such as Tris and glycine—as well as thiol‑containing compounds must be avoided. Ammonium salts commonly used for protein precipitation, such as ammonium sulfate and ammonium acetate, should also be removed prior to dye conjugation, for example by dialysis. 4. Reaction temperature: Most conjugation reactions proceed at room temperature. However, certain labeling reactions may require elevated or reduced temperatures.
