Transportation and Storage
Store at room temperature, protected from light. Shelf life: 12 months.
Instructions for Use
1. For adherent cells (using a 6-well plate as an example)
(1) Culture cells in a 6-well plate.
(2) Place the 6-well plate on ice and rinse the cells twice with pre-cooled 4°C PBS.
(3) Fix the cells in pre-cooled 100% methanol at 4°C for 10 minutes. Ensure that the methanol completely covers the cells to achieve uniform fixation.
(4) Aspirate the methanol from the well plate. If you need to temporarily halt the experiment, add PBS containing 50% glycerol to cover the cells. Seal the 6-well plate with sealing film or plastic wrap and store it at 4°C; it can be kept for up to one week.
(5) Remove the cells from the freezer, allow them to equilibrate to room temperature, then add an appropriate amount of crystal violet staining solution to cover the cells. Incubate at room temperature for 10 minutes, discard the crystal violet stain, and the staining solution can be reused.
(6) Rinse the cells repeatedly with distilled water until no residual crystal violet remains in the wash solution. Ensure that excess dye is thoroughly removed to prevent background interference.
(7) Microscopic examination.
2. For suspension cells: the same procedures as for adherent cells may be followed.
Precautions

2. For your safety and health, please wear a lab coat and disposable gloves while performing this procedure.
3. When pipetting the staining solution, avoid contact with the cells.
4. When using this kit for the first time, it is recommended to perform a preliminary experiment with 1–2 samples.
