Product Description
Romanowsky stain is a composite dye composed of the acidic dye eosin and the basic dye methylene blue. Its staining characteristics are as follows: hemoglobin and acidophilic granules, which are basic proteins, bind to the acidic dye eosin and appear pink; nuclear proteins, which are acidic, bind to the basic dyes methylene blue or azure and stain purplish‑blue; and neutral granules, being at the isoelectric point, can bind to both eosin and methylene blue, resulting in a pale purple color.
Giemsa stain consists of azure and eosin; its staining mechanism is similar to that of the Wright stain, though it exhibits weaker staining for cytoplasm and neutral granules. To combine the advantages of both methods, the Wright–Giemsa composite stain is recommended, as it provides a more comprehensive visualization of cellular structures and components.

Instructions for Use
1. Smear and fix: Spread the cells evenly onto a clean microscope slide, allow them to dry, and then proceed with fixation.
Note: The choice of fixative depends on the specific experimental conditions; methanol is suitable for most cell types. Methanol fixation: After the smear has dried, immerse it in methanol for 15–30 minutes.
2. Dyeing:
(1) After fixation, allow the slides to air-dry at room temperature. Place the slides in the staining tank and prepare for staining.
(2) Pour Wright–Giemsa stain into the staining tank, ensuring that the stain fully covers the cells on the slide. The specific staining time depends on the cell type; typically, it ranges from 3 to 30 minutes. During staining, it is advisable to observe the preparation under a microscope.
(3) After staining, perform a water wash: first rinse the slides with PBS, then gently rinse with a thin stream of water to prevent large numbers of cells from being washed off the slide and compromising subsequent observations. Once rinsing is complete, place the slides in a cool, shaded area to air-dry.
3. Mounting: Mount with neutral gum arabic to prepare a permanent slide.
4. Microscopic observation: Place the dried slide under the microscope for examination.
Staining results: The cell nuclei are stained blue, while the cytoplasm is stained red.
Precautions
1. This product is intended solely for scientific research and must not be used for clinical diagnosis, treatment, or any other purposes.
2. For your safety and health, please wear a lab coat and disposable gloves when performing this procedure.
3. Staining time depends on room temperature and the number of cells. At lower temperatures or with a higher cell density, longer staining times are required; conversely, shorter times may be used. If necessary, increase the volume of staining solution or extend the incubation period. Before rinsing, first examine under low magnification to confirm that nuclear staining is clear and that nuclear–cytoplasmic contrast is distinct. The optimal staining duration should be determined through practical experimentation, particularly when changing the staining solution. Each batch of staining solution must be tested to establish the appropriate staining time.
4. During rinsing, do not discard the staining solution first; instead, rinse thoroughly with running water, as failure to do so may cause the dye to precipitate on the slide.
5. After rinsing, the slides should be placed upright on a rack to prevent residual water from soaking and causing decolorization.
6. When using this kit for the first time, it is recommended to perform a preliminary experiment with 1–2 samples.
