Product Description
Giemsa stain consists of azure and eosin, and its staining principle is essentially the same as that of Wright’s stain. Specifically: acidophilic granules, which are basic proteins, bind to the acidic dye eosin, resulting in a pink color; nuclear proteins and lymphocyte cytoplasm, being acidic, bind to the basic dyes methylene blue or azure, yielding a purplish‑blue hue; and neutral granules, existing in an isoelectric state, can bind to both eosin and methylene blue, producing a pale purple color.
The staining outcome is influenced by pH. All cellular components contain proteins, which are amphoteric electrolytes; their net charge depends on the solution’s pH. In slightly acidic conditions, the positive charge increases, facilitating binding with eosin and yielding a reddish stain; in slightly alkaline conditions, the negative charge predominates, promoting binding with azure and resulting in a bluish hue. Consequently, cellular staining is highly sensitive to hydrogen ion concentration. Slides used for staining must be thoroughly cleaned, and the rinsing water should be near neutral; otherwise, abnormal staining reactions may occur, making cell identification challenging.

Transportation and Storage
Store at room temperature, protected from light. Shelf life: 12 months.
Instructions for Use
1. Reagent preparation: Dilute the Giemsa stock solution 10-fold with 0.1 M PBS to obtain the working Giemsa solution.
2. Smear: Prepare a smear with uniform cell density; alternatively, directly dispense a cell suspension of adjusted density onto the slide and allow it to air-dry promptly.
3. Fixation: First fix with Giemsa working solution for about 1 minute, then add PBS; the ratio of the two should not exceed 1:1. Be sure to keep the slide moist before adding PBS; otherwise, excessive sediment will form, leading to uneven staining.
4. Staining: The specific staining time depends on the cell type; typically, it ranges from 3 to 30 minutes. It is best to observe the staining process under a microscope.
5. Washing: During rinsing, carefully control the water flow—too little may leave fine dye residues adhering to the slide, while too much could exacerbate delamination. Slides that have been stained can also be further refined and corrected. If the staining is too light, a second stain can be applied; if it is too dark, brief destaining with ethanol may be used.
6. Microscopic examination: observed under a microscope.
Staining results: The cell nuclei are stained purplish-red or purplish-blue, while the cytoplasm is stained light red.
Precautions
1. This product is intended solely for scientific research and must not be used for clinical diagnosis, treatment, or any other purposes.
2. For your safety and health, please wear a lab coat and disposable gloves when performing this procedure.
3. The staining working solution should be prepared fresh as needed; aged staining solutions yield poor results. The working solution should not be stored for more than 48 hours.
4. Giemsa stain is highly sensitive to pH; therefore, the pH of the PBS must be precisely controlled, or the staining result will be suboptimal. When using a staining jar, an oxidized film (which appears shiny) often forms on the surface of the staining solution as the incubation time increases. This film can adhere to the slide, causing contamination that is difficult to remove—particularly when non‑freshly prepared reagents are used. Before staining, gently scrape off the oxidized layer from the liquid surface with a small piece of filter paper, then proceed with staining. After staining is complete, immerse the specimen in water to rinse away any residual stain.
5. When using this kit for the first time, it is recommended to perform a preliminary experiment with 1–2 samples.
