Product Description
Propidium iodide (PI) is a widely used fluorescent nuclear stain, analogous to ethidium bromide (EB). It intercalates between DNA base pairs without significant sequence preference, binding approximately one dye molecule every 4–5 base pairs. PI can also bind to RNA; to distinguish between DNA and RNA staining, nuclease treatment is typically required. The maximum excitation and emission wavelengths of aqueous PI solutions are 493 nm and 636 nm, respectively; however, upon binding to nucleic acids, the fluorescence signal is enhanced by 20–30-fold, with the peak excitation and emission wavelengths shifting to 535 nm and 617 nm. PI is suitable for analysis using fluorescence microscopy, confocal microscopy, flow cytometry, and fluorometers.
PI cannot penetrate the cell membrane and is therefore excluded from living cells, but it can pass through damaged membranes and stain nuclei. Leveraging this property, PI is commonly used in combination with live-cell fluorescent probes such as Calcein‑AM, Hoechst 33258 (Cat. No.: AC12L012), or Hoechst 33342 (Cat. No.: AC12L022) to simultaneously stain and distinguish between live and dead cells, making it suitable for studies related to apoptosis. Additionally, PI serves as a counterstain in multiplex fluorescence assays and is compatible with a wide range of cell‑labeling techniques, including direct or indirect fluorescent antibody detection, mRNA in situ hybridization, cell‑structure‑specific fluorescent probe–based assays, and tissue staining. PI staining is also well suited for cell‑cycle analysis.
This product is a PI stock solution in aqueous form, with a concentration of 1 mg/mL; dilute to the appropriate working concentration before use.
