Product Description
Ham's F-12 The Ham’s F‑12 Nutrient Mixture was developed by Ham in 1969, based on the earlier Ham’s F‑10 formulation, and was initially designed for serum‑free culture of CHO cells. Ham’s F‑12 is commonly used as a basal medium for serum‑free cultures; at low serum concentrations, it is particularly well suited for single‑cell and clonal cultures. When supplemented with serum, it is also widely employed for culturing cancer cell lines and primary cells, including rat hepatocytes, rat prostatic epithelial cells, chondrocytes, rat myoblasts, and chicken embryo cells. Furthermore, mixing Ham’s F‑12 with DMEM in equal volumes yields a DMEM/F‑12 medium that is nutritionally richer and more extensively utilized.
Compared with other basal media, F-12 contains a broader range of components, including zinc, putrescine, hypoxanthine, and thymidine. F-12 does not contain proteins or growth factors; therefore, it requires supplementation, typically with 10%. Fetal Bovine Serum, Premium Grade (Catalog No.: AC03L055) . F-12 uses a sodium bicarbonate buffering system (1.176 g/L), thus requiring a 5–10% CO2 environment to maintain physiological pH.
