Ham's F-12K (Kaighn's) The culture medium is an improved formulation of Ham’s F-12 Nutrient Mixture. Ham’s F-12K (Kaighn’s) medium was specifically developed for primary human hepatocytes and for certain rat and chicken hepatocytes cultured under serum-reduced conditions.
Ham's F-12K (Kaighn's) The culture medium contains a variety of components not found in conventional basal media, such as putrescine, thymidine, hypoxanthine, zinc, and elevated levels of all amino acids and sodium pyruvate. With these additions, certain cell types require only minimal supplementation with serum or well-defined components. Ham’s F‑12K (Kaighn’s) medium is devoid of proteins or growth factors; therefore, it is typically supplemented with growth factors and fetal bovine serum (FBS). The optimal FBS concentration must be determined for each specific cell line. Ham’s F‑12K (Kaighn’s) medium employs a bicarbonate buffering system (2.5 g/L), necessitating an atmosphere of 5–10% CO₂ to maintain physiological pH.
