Product Description
Phalloidin is a bicyclic heptapeptide toxin that specifically binds to the interface between F‑actin subunits, locking adjacent subunits together. Compared with actin monomers, phalloidin exhibits a much stronger affinity for actin filaments; by reducing the rate constant for the dissociation of actin subunits from filament ends, it stabilizes actin filaments and thereby inhibits filament depolymerization. Furthermore, studies have shown that phalloidin can also inhibit the ATPase activity of F‑actin. This toxin stabilizes F‑actin structures by trapping actin monomers in a conformation distinct from that of G‑actin and by markedly decreasing the rate constant for monomer dissociation associated with ADP binding.
The effects of phalloidin in cells vary with its concentration. At low concentrations, it recruits low‑polymerization‑state actin and filamin into stable actin polymer “islands” without disrupting the structure of stress fibers—i.e., bundled microfilaments. This property makes phalloidin a widely used tool for studying F‑actin distribution: fluorescently labeled phalloidin derivatives can be used to stain actin filaments for observation by light microscopy. Fluorescent phalloidin derivatives not only enable precise localization of actin filaments in live or fixed cells but also facilitate the visualization of individual actin filaments in vitro, serving as a critical instrument for high‑resolution investigations of the actin cytoskeleton.
This product is characterized by high binding specificity, selectively binding to polymerized F‑actin (filamentous actin) while exhibiting minimal binding to monomeric G‑actin, resulting in a high signal‑to‑noise ratio. Applicable sample types: only for fixed cells—fixed and permeabilized cells, cell cultures, cell‑free assays, and formaldehyde‑fixed and permeabilized tissue sections.
