Product Description
Phalloidin‑AMCA specifically binds to F‑actin. At nanomolar concentrations, this derivative can be used to label, identify, and quantitatively analyze F‑actin in formaldehyde‑fixed and permeabilized tissue sections, cell cultures, or cell‑free assay systems. Compared with actin monomers, phalloidin exhibits a higher binding affinity for actin filaments and markedly reduces the rate constant for the dissociation of actin subunits from filament ends, thereby stabilizing actin filaments by inhibiting filament depolymerization. Studies have also shown that this compound can suppress the ATPase activity of F‑actin.
The functional effects of phalloidin in cells are concentration‑dependent: at low concentrations, when applied to the cytoplasm, it recruits monomeric cytosolic actin and filamin into stable actin polymer “islands” without disrupting the architecture of stress fibers (i.e., bundled microfilaments). Staining actin filaments with fluorescently labeled phalloidin derivatives provides an effective approach for investigating the intracellular distribution of F‑actin. Fluorescent phalloidin conjugates not only enable precise localization of actin filaments in live or fixed cells but also allow visualization of individual actin filaments in vitro, making them an indispensable tool for high‑resolution studies of the actin cytoskeleton.
This product features With its highly specific binding properties, it selectively binds to F‑actin (filamentous actin) while exhibiting minimal interaction with G‑actin (monomeric actin), enabling clear visualization of the cytoskeleton’s fine architecture with low background interference and crisp imaging. Applicable sample types: exclusively for fixed cells—fixed and permeabilized cells, cell cultures, cell‑free assays, and formaldehyde‑fixed, permeabilized tissue sections.
