Product Description
Phalloidin–fluorescein isothiocyanate conjugates (equivalent to FITC‑labeled phalloidin) selectively bind to F‑actin. At nanomolar concentrations, these phalloidin derivatives serve as research tools for labeling, detecting, and analyzing F‑actin in formaldehyde‑fixed and permeabilized tissue sections, cell cultures, or cell‑free assay systems.
Phalloidin exhibits a higher binding affinity for filamentous actin than for actin monomers, and its primary mechanisms of action include: 1) decreasing the rate constant for the dissociation of actin subunits from the filament ends; 2) stabilizing the actin filament structure by inhibiting filament depolymerization; and 3) suppressing the ATPase activity of F‑actin.
Phalloidin exerts concentration‑dependent effects within cells. At low concentrations, it recruits low‑polymerization‑state actin and filamin into stable actin polymer structures without disrupting the integrity of stress fibers—i.e., bundled microfilaments. Owing to these properties, phalloidin serves as a valuable tool for investigating the subcellular distribution of F‑actin; fluorescently labeled phalloidin enables staining of actin filaments for visualization by light microscopy. Fluorescently tagged phalloidin derivatives are highly useful in basic biomedical research, allowing the localization of actin filaments in fixed cells, the direct visualization of individual actin filaments in vitro, and high‑resolution imaging of actin networks; they have already found widespread application in the study of the cytoskeleton.
This product exhibits specific binding characteristics, selectively associating with F‑actin (filamentous actin) while showing minimal interaction with G‑actin (monomeric actin), making it suitable for fluorescence imaging studies of the cytoskeleton’s fine architecture. It supports a variety of fluorescent labels and can be conjugated with diverse fluorophores—including FITC, TRITC, the Alexa Fluor® series, AMCA, Cy dyes, and others—covering wavelengths from the ultraviolet to the near‑infrared to meet multi‑color imaging requirements. It offers low background interference and produces sharp, high‑contrast images. Applicable sample types: exclusively for fixed cells; fixed and permeabilized cells; cell cultures; cell‑free assays; and formaldehyde‑fixed, permeabilized tissue sections.
