Product Description
Phalloidin–rhodamine conjugates (equivalent to TRITC‑labeled phalloidin) selectively bind F‑actin and exhibit markedly superior photostability compared with fluorescein (FITC)‑labeled phalloidin conjugates. At nanomolar concentrations, these phalloidin derivatives serve as convenient probes for labeling, visualizing, and quantifying F‑actin in formaldehyde‑fixed and permeabilized tissue sections, cell cultures, or cell‑free assay systems. Phalloidin’s binding affinity for actin filaments is significantly higher than that for monomeric actin; its mechanism of action primarily involves: 1) a substantial reduction in the rate constant for the dissociation of actin subunits from filament ends; 2) stabilization of actin filament architecture by inhibiting filament depolymerization; and 3) suppression of ATP hydrolysis activity in F‑actin.
The effects of phalloidin in cells are concentration‑dependent. At low concentrations, it recruits poorly polymerized actin and filamin from the cytoplasm to form stable actin‑polymer “islands,” while leaving the structure of stress fibers (i.e., bundled microfilaments) unaffected. This property makes phalloidin an invaluable tool for studying the intracellular distribution of F‑actin: by fluorescently labeling phalloidin and staining actin filaments, one can visualize them using light microscopy.
Fluorescent phalloidin derivatives have demonstrated significant value in biomedical research: 1) they enable precise localization of actin filaments in live and fixed cells; 2) they allow high‑resolution visualization of individual actin filaments in vitro; and 3) they are well suited for studying actin networks at high resolution. These derivatives have been widely employed in cytoskeletal research.
This product exhibits highly specific binding, selectively interacting with F‑actin (filamentous actin) while showing minimal binding to G‑actin (monomeric actin), enabling clear visualization of the cytoskeleton’s fine architecture. It features low background interference and delivers sharp imaging. Applicable sample types: exclusively for fixed cells—fixed and permeabilized cells, cell cultures, cell‑free assays, and formaldehyde‑fixed, permeabilized tissue sections.
