Product Description
Collagenase is a crude enzyme preparation derived from Clostridium histolyticum, also known as clostridiopeptidase A. It contains not only collagenase but also a mixture of several other enzymes that work together to dissociate cells and tissues. This “crude” collagenase preferentially degrades the helical regions of native collagen and can cleave multiple times within the triple‑helix structure, targeting the Y–Gly bond in the Pro–Y–Gly–Pro sequence—common in collagen and the principal fibrous component of extracellular connective tissue in animals—where Y is most often a neutral amino acid. Compared with trypsin, collagenase exhibits milder digestion, maintaining excellent dissociation activity under physiological temperature and pH conditions without the need for mechanical agitation.
Currently, commercially available bacterial collagenases are classified into four types based on differences in collagenase activity: Type I, Type II, Type III, and Type IV, each with specific applications: (1) Type I Collagenase contains a relatively balanced spectrum of enzymatic activities, including collagenase, caseinase, clostridial protease, and trypsin. It is commonly used for the isolation of epithelial cells, as well as cells derived from liver, lung, adipose tissue, and adrenal gland tissues. (2) Type II Collagenase exhibits higher clostridial protease activity and is typically employed for isolating cells from tissues such as heart, bone, muscle, thymus, and cartilage. (3) Type III Collagenase has lower protease activity and is frequently used for the preparation of mammary gland cells. (4) Type IV Collagenase features low trypsin activity and is generally utilized for isolating pancreatic islet cells or for experiments requiring the preservation of membrane proteins or receptors.
